What Is HPLC Peptide Testing? A Buyer’s Guide
When you buy a research peptide, the single most important number is purity, and purity is not something you can see, smell, or guess. It has to be measured. The standard tool for that measurement is HPLC, High-Performance Liquid Chromatography. If you have ever looked at a certificate of analysis and seen a figure like “98% by HPLC,” this is where that number comes from. This guide explains what HPLC testing is and how to read it as a buyer.
What HPLC actually does
HPLC is an analytical technique that separates the components of a sample so they can be measured individually. A peptide sample is dissolved and pushed under high pressure through a column packed with a specialised material. Different molecules travel through that column at different speeds depending on their chemistry, so they come out separated in time.
A detector at the end records each component as a peak on a chromatogram, a graph of signal against time. The main peptide shows up as one large peak; impurities show up as smaller peaks. By comparing the area of the main peak to the total, the lab calculates a purity percentage. That is the number that ends up on the certificate.
Why purity matters
In research, consistency is everything. If a peptide sample is only partly the intended compound, the rest is impurities, truncated sequences, byproducts, or residual solvents, then results become harder to reproduce and compare. Two batches at different purities are effectively two different inputs. A verified purity figure is what lets a researcher trust that this lot behaves like the last one.
What HPLC does and does not tell you
HPLC is excellent at answering “how pure,” but on its own it does not confirm “is this the right molecule.” That is why serious testing pairs HPLC with mass spectrometry (MS):

How to read a certificate of analysis
A certificate of analysis (COA) is the document that reports these results for a specific lot. When you review one, look for:
- Purity percentage by HPLC, and ideally the chromatogram itself.
- Identity confirmation by mass spectrometry, showing the expected molecular mass.
- Lot or batch number, so the COA is tied to the actual product you receive, not a generic sample.
A COA that matches the lot in your hands is far more meaningful than a single test result posted once and reused for everything. Lot-specific testing is the standard worth insisting on.
