Growth Hormone Peptides: A Research Overview

Growth hormone peptides are one of the most researched families in the field, and three names come up more than any others: sermorelin, CJC-1295 and ipamorelin. They are often grouped together, but they are not interchangeable. This overview explains what each one is, how they differ, and why two of them are so frequently paired.

The core idea: stimulate, do not replace

Unlike synthetic growth hormone itself, these peptides do not supply GH directly. They are secretagogues, meaning they signal the pituitary gland to release its own growth hormone. That distinction is the reason the category exists as a separate research area, and it is why timing and pulsatility feature heavily in the literature.

Sermorelin

Sermorelin is a truncated analogue of growth hormone-releasing hormone (GHRH), corresponding to the first 29 amino acids that carry the biological activity. It binds the GHRH receptor on the pituitary. Its defining characteristic is a very short half-life, which produces a brief, sharp signal. In research terms it is the classic, first-generation GHRH analogue that the newer compounds were designed to improve on.

CJC-1295

CJC-1295 is also a GHRH analogue, but it is engineered for a longer duration of action. The version LYFE carries is CJC-1295 (No DAC), sometimes called modified GRF (1-29). The “No DAC” label matters: the DAC (Drug Affinity Complex) variant binds albumin for an extended half-life measured in days, while the No DAC version gives a shorter, more pulse-like signal that pairs cleanly with a ghrelin mimetic. Both are GHRH analogues; they differ mainly in how long the signal lasts.

Ipamorelin

Ipamorelin sits in a different class. It is a growth hormone-releasing peptide (GHRP) and a selective ghrelin-receptor agonist. Because it works through a separate pathway from GHRH, it is prized in research for its selectivity: it is studied as one of the cleaner GHRPs, with less spillover onto other hormonal pathways than older compounds in its class.

Why CJC-1295 and ipamorelin are paired

The pairing is the whole point. GHRH analogues (CJC-1295) and ghrelin mimetics (ipamorelin) act on two different receptors, and the research interest lies in how the two signals combine. Using one from each family is a standard design in the literature precisely because they probe complementary pathways rather than the same one twice.

Why selectivity matters

One reason ipamorelin gets so much attention is its clean profile. Earlier ghrelin-receptor peptides in the same class are known for pushing on other pathways at the same time, for example strong appetite signalling or shifts in cortisol and prolactin. Ipamorelin is studied as a more selective option that concentrates its activity on the growth-hormone pathway with fewer of those off-target effects. In a research context, a more selective tool means fewer confounding variables, which is exactly what makes it a preferred pairing partner for a GHRH analogue.

How to think about the three

Sermorelin and CJC-1295 are relatives (both GHRH analogues), separated mainly by how long they act. Ipamorelin is the outsider, working through the ghrelin pathway instead. That is why the most common research configuration is a GHRH analogue plus ipamorelin rather than two GHRH analogues together.
